산양유의 산업적 이용 가치에 대한 연구 고찰

Abstract

In many countries, goat milk is an excellent nutrient source and is less allergenic for children and the elderly. The casein composition of goat milk consists largely of β-casein and lower amounts of αs1-casein, which may interfere with digestion by forming solid curds in the human stomach. Goat milk contains small fat globules and large amounts of medium chain fatty acids for, better digestibility, as well as abundant minerals and vitamins with high absorption rates. Recently, the medical benefits of goat milk in different human disorders have been recognized, leading to an increased interest in developing functional foods with goat milk, particularly for individuals with malabsorption syndrome. However, the physiological and biochemical properties of goat milk are largely unknown. We review the importance of goat milk as a potential functional food by providing scientific evidence confirming its health benefits.

서 론

포유동물의 젖은 복잡한 화학적, 물리적 특성을 가지고 있으며, 이러한 특성의 차이는 종 (species), 유전적 요인, 환경 조건 등에 영향을 받는다(Kanwal et al., 2004). 특히, 사람의 모유는 영아의 건강한 성장 및 발달에 가장 이상적인 식품으로 다양한 생리활성 물질들이 포함되어 있을 뿐 아니라, 유해한 병원균의 침입을 방어하는 역할을 한다(Sobti et al., 2002; Martinez-Ferez et al., 2006; Hilton, 2013). 우유 (cow milk)는 인체에 유익한 영양소들의 우수한 공급원으로 모유 수유를 할 수 없는 영아들에게 대체할 수 있는 유일한 식품으로 인식되어 왔다. 그런데 최근 들어 인간이 섭취하는 다양한 포유 동물의 젖들 중에서 모유의 성분 및 조성에 유사한 특성을 보유한 산양유가 주목을 받게 되었으며, 기능성 식품 시장이 활발하게 성장하면서 산양유의 영양학적인 이점과 저알러지 특성으로 인해 그 활용성이 증가되어 다양한 나라에서 건강한 이미지의 식품 소재로 자리 잡고 있다(Haenlein, 2004; Raynal-Ljutovac et al., 2005; Siro et al., 2008; Albenzio and Santillo, 2011; Yangilar, 2013).

산양유는 케이신(casein)의 한 종류인 αs1-케이신의 함량이 적어 부드러운 커드를 형성하고, 단백질 및 지방산 구성이 모유와 유사하기 때문에 소화 흡수율이 뛰어나, 산양유와 그 가공 제품은 특히 영유아 및 노인에게 건강 유지를 위한 적합한 식품일 뿐 아니라, 알레르기에 민감한 환자들에게는 기능성 식품으로 유용하게 활용될 수 있다(Jandal, 1996; Park, 2000; Bramanti et al., 2003).

이러한 산양유의 특성으로 인해 전 세계적으로 산양유의 생산비율은 다른 포유동물과 비교하여 크게 증가하고 있으며, 2010년에 산양유와 관련된 유제품을 소비하는 인구는 약 1억 9천만명에 도달하였음을 알 수 있다(Haenlein, 2004; Talpur et al., 2009; Faostat, 2012). 산양유에 대한 수요는 아시아, 중동, 남아메리카, 유럽, 북아메리카, 오세아니아 지역 등의 개발도상국 및 선진국들에서 시장규모 확대와 함께 증가하고 있어 경제적 중요성이 부각되고 있다(Albenzio et al., 2006). 특히, 개발도상국에서는 공식적인 통계 수치로 보고되는 산양유의 시장 규모보다 보고되지 않은 가정에서의 산양유 소비 규모가 더 커서 이용 범위는 더욱 넓을 것이라 추정된다(Yangilar, 2013).

우리나라의 경우, 산양유를 이용한 분유제품이 출시된 2003년 이후 산양유 시장이 본격적으로 시작되었으며 특히, 2011년 구제역 사태 이후로 다양한 종의 가축을 사육해야 한다는 필요성이 부각되면서 질병에 강하고 젖소에 비하여 체중 대비 산유량이 2배가 높은 유산양을 사육할 수 있는 환경을 조성해야 한다는 주장이 제기되기도 하였다(Ahn and Park, 2008).

이에 본 총설은 산양유의 시장 요구도에 발맞춰 현재까지 알려진 산양유의 이화학적, 영양학적 특성 및 생리활성에 관한 연구 결과에 대해 정리 및 고찰함으로써 앞으로 산양유의 연구방향 설정과 활용방안에 도움이 되고자 수행되었다.

본 론

1. 산양유의 일반 성분과 이화학적 특성

산양유의 일반 영양성분 조성 및 함량은 산양의 사료 조건, 분만 시기, 계절, 환경, 사육 지역, 건강상태 등의 조건에 따라 변동될 수 있으며, 평균함량은 Table 1과 같다. 일반적으로 산양유의 영양성분 조성은 우유와 유사하며, 연구자와 지역에 따라 다소 차이를 보인다. 지방의 경우, 우유보다 다소 높은 함량을 보이지만, 산양유의 지방구 크기는 더 작아 식품 제조 시에 부드러운 질감을 제공하며, 콜레스테롤의 함량이 낮은 것이 특징이다(Keskin et al., 2004; Küçükçetin et al., 2011; Toral et al., 2015). 유당의 함량은 모유와 우유에 비해 대체로 낮은 반면 회분의 경우는 높은 경향을 보인다.

Attaie와 Richter(2000)의 연구에 따르면 산양유는 렌넷 처리에 의한 응고시간이 길며, 위산으로 인해 응고된 커드가 부드럽고 연약하여 인체 내에서 가수분해 및 소화 흡수가 용이하다. 산양유 케이신 마이셀의 직경은 약 80 nm로 평균 100~250 nm인 우유의 마이셀 직경보다 작아 공간 구조적 차이가 있고, 케이신 단백질의 아미노산 조성 등에도 명백한 차이를 보인다(Kim et al., 2014).

산양유에는 다양한 생물학적 기능성을 가진 타우린과 높은 항산화, 항암효과 및 면역기능에 관여하는 셀레늄, 그리고 항암작용, 면역증진 효과 및 체지방 감소 기능을 가진 CLA(conjugated linoleic acid) 등이 포함되어 있다(Manzi and Pizzoferrato, 2013; Toral et al., 2015). 또한, 산양유는 장내 미생물내 유익균의 성장과 활성을 선택적으로 증진시켜 숙주 건강을 개선시키는 올리고당의 함량이 모유보다는 적게 함유되어 있지만, 우유에 비해 약 4~10배 정도 풍부하다(Table 1).

2. 산양유의 영양적 특성
1) 단백질

산양유의 주요 단백질 조성은 αs-케이신, β-케이신, κ-케이신, α-락트알부민, β-락토글로불린으로 우유와 비슷하지만, 각 케이신의 함량은 큰 차이를 보인다(Table 2). αs1-케이신은 소화과정에서 위산에 의해 응고되어 다소 단단한 커드를 형성하기 때문에 소화 흡수에 방해 요인이 될 가능성이 있는데, 산양유 케이신은 β-케이신의 함량이 높고 αs1-케이신의 함량은 낮아 부드러운 커드를 형성하여 소화 흡수가 용이하다(Remeuf and Lenoir, 1986; Renner et al., 1989; Park and Haenlein, 2006; Tomotake et al., 2006). 이러한 특성은 산양유의 우수한 소화, 흡수 능력과도 관련이 있을 것이라 판단된다(Richardson and Creamer, 1975). 모유의 경우, αs-케이신은 발견되지 않으며, 주된 성분은 β-케이신으로 알려져 있다. 특히, 산양유 케이신의 분자 구조는 모유와 유사하여 소화, 흡수력이 좋기 때문에 복통이나 설사 등의 증상이 나타날 가능성이 낮다. 또한, 임상연구에서도 산양유가 알러지 반응을 야기하는 확률이 낮아, 영유아나 노인뿐만 아니라, 소화에 어려움을 겪는 일부 사람들에게 우수한 영양 공급원이 될 수 있다(Jenness, 1980; Restani et al., 1999).

유단백질 중 β-케이신의 유전적 변이체들은 A1, A2, A3, B, C, D, E, F, H1, H2, I, G 등으로 다양하지만, A1 및 A2 변이체가 가장 일반적인 형태이며, 산양유의 β-케이신은 주로 A2 변이체로만 구성되어 있다(Cieślińska et al., 2012). β-케이신은 총 209개의 아미노산 서열로 구성되어 있는데, A1 변이체에는 아미노산 서열 67번째에 히스티딘이 포함되어 있고, A2 변이체에는 프롤린이 포함되어 있다(Farrell et al., 2004). A1 변이체의 히스티딘은 체내에서 가수분해되어 β-카소모르핀 7(Tyr-Pro-Phe-Pro-Gly-Pro-Ile)을 생성하지만, A2 변이체는 이러한 단편이 생성되지 않는 것이 특징이다. β-카소모르핀 7은 중추신경 및 위장관 내의 μ-오피오이드 수용체와 결합하며, 잠재적인 병인성 요인이 될 가능성이 있다고 알려져 있지만, A2 변이체는 어떠한 질병 발생과 무관한 것으로 보고되어 향후 지속적인 연구가 필요한 실정이다(Teschemacher, 2003; Kostyra et al., 2004; Cieślińska et al., 2012).

미국 농무부에서 제시한 산양유와 우유의 필수 및 비필수 아미노산의 평균 함량은 Table 3과 같다(Davis et al., 1994; Park and Haenlein, 2006). 모유와 우유와 비교할 때 산양유에는 트레오닌과 발린 등의 필수아미노산이 높게 함유되어 있으나, 아직 아미노산 조성 차이에 따른 대사 효과에 관한 연구결과는 부족한 실정이다(Haenlein, 2004). 그러나 흰쥐를 이용한 소화 흡수 연구에서 산양유 특유의 필수아미노산 구성에 의해 구리의 장내 소화 흡수력이 증진된다고 보고된 바 있다(Barrionuevo et al., 2002; Yangilar, 2013).

2) 지방

산양유 내 지방산의 평균 함량은 모유나 우유와 비교하여 차이를 보인다(Table 4). 특히, 산양유의 지방산 중 카프로산(C6:0), 카프릴산(C8:0), 카프릭산(C10:0), 라우르산(C12:0), 미리스트산(C14:0)의 함량은 모유나 우유보다 높다. 산양유 지질의 중요한 특성은 건강에 유익하다고 알려진 중간사슬지방산의 함량이 높다는 것으로 카프릴산과 카프릭산 등의 중간사슬지방산은 지방조직에 축적되지 않고, 직접 에너지로 전환되는 특별한 대사 과정을 거친다(Haenlein, 2004). 이러한 지방산 조성은 혈청 콜레스테롤 수치 저하 및 콜레스테롤 축적을 억제하거나 제한하여 대사증후군, 유미뇨증, 지방변증, 고리포단백혈증, 유아영양실조, 간질, 낭포성섬유증, 담석 등의 질환에 대한 치료제로 인정받고 있다(Haenlein, 2004; Kompan and Komprej, 2012).

일반적으로 지질의 소화율은 지질에서 유리되는 유리지방산의 함량으로 결정되는데, 산양유 및 우유의 지질 소화율은 각각 0.62 mEq/L와 0.12 mEq/L로 우유보다 높았으며, 리파아제를 처리한 실험에서도 산양유의 지질이 잘 분해되었다(Strzalkowska et al., 2009; Kim et al., 2014). 이러한 특성들은 아직 산양유 유제품 판매에 마케팅 전략으로 사용되고 있지는 않지만, 산양유는 훌륭한 영양공급원인 동시에 의학적으로도 매우 큰 잠재성을 가지고 있다는 것을 알 수 있다(Babayan, 1981; Haenlein, 2004).

산양유 내 지방산 조성은 산양을 사육할 때 공급하는 조섬유 조성의 비율에 따라 변화될 수 있다. 목초 사료를 높은 비율로 섭취한 산양유는 부티르산, 카프로산, 스테아르산, 올레산, 리놀렌산, 아라키돈산의 함량은 높고, 카프릭산, 라우르산, 미리스트산, 팔미트산, 리놀레산의 함량은 낮았으며, 비타민 A, 토코페롤, 미네랄 등을 다량 함유하고 있는 알팔파를 원재료로 하는 사료를 높은 비율로 섭취한 경우, 산양유 내 유익하지 않은 트랜스 올레산의 함량이 매우 낮았다(Alonso et al., 1999; LeDoux et al., 2002). 또한 사료 조성을 적절하게 조절하여 공급하면 유익한 불포화지방산 함량을 높일 수 있고, 산양유의 영양학적 가치를 상승시킬 수 있다고 보고되었다(Sanz Sampelayo et al., 2002; Haenlein, 2004).

3) 비타민과 무기질

산양유와 우유의 비타민 및 무기질 함량은 Table 5, 6과 같다. 비타민의 경우, 산양유는 모유와 우유에 비해 티아민, 나이아신, 피리독신 등의 함량이 높았으며, 무기질의 경우, 칼슘, 인, 칼륨, 염소, 마그네슘, 망간 등의 함량이 높게 함유되어 있다. 그러나 비타민과 무기질의 경우, 함량의 차이보다는 각 영양소의 이용성이 더 중요하며, Rutherfurd 등(2006)의 연구에 따르면 산양유의 칼슘, 인, 망간은 체내 흡수율 및 이용률이 뛰어나다고 보고하였다. 이러한 특성으로 인해 다양한 형태의 치료식이나 건강식이 개발되어 그 이용성이 증가되고 있는 추세이다(Attaie and Richter, 2000; Mowlem, 2005). 반면, 또 다른 보고에 따르면 산양유 내 코발라민과 엽산의 함량은 낮아 영유아의 거대적 아구성 빈혈과 엽산 결핍성 빈혈에 취약하기 때문에 다른 식품섭취 또는 가공을 통하여 보완할 필요성이 있다고 강조한 바도 있다(Park et al., 2007; Bernacka, 2011).

3. 산양유의 기능성

산양유가 생리 활성에 유익한 영향을 미치는 것은 많은 선행연구를 통하여 알 수 있다(Parkash and Jenness, 1968; Ahn and Park, 2008). 산양유를 섭취한 쥐의 혈액을 채취하여 분석한 결과, 노화나 혈관 장애 정도를 알 수 있는 과산화지질 수준이 다른 유를 섭취한 쥐보다 낮은 것을 확인할 수 있었고, 발효한 산양유를 섭취한 사람들에게 항산화 효과에 이로운 영향을 미치는 것으로 나타났다(Silva and Malcata, 2005; Korhonen and Pihlanto, 2006; Madureira et al., 2007; Phelan et al., 2009; Díaz-Castro et al., 2012). 산양유 유래 생리 활성 펩타이드는 항산화, 항균작용 및 면역조절 작용에 더불어 항혈전 작용을 통한 심혈관계 질환 방지 효과가 보고되었으며, 체액량 조절이나 혈압에 있어 중요한 역할을 하는 안지오텐신 전환 효소(Angiotensin Converting Enzyme) 저해 작용에도 관여한다는 것이 보고되었다(Quirós et al., 2005; Eriksen et al., 2008; Atanasova and Ivanova, 2010).

Oh 등(2016)의 연구에서 산양유 발효물은 면역학적 활성과도 관련이 있으며, 발효 산양유를 경구 투여한 마우스는 T세포와 B세포 등의 림프구가 활성화되어 ConA 및 LPS와 같은 미토겐(mitogen)에 대한 반응성이 높아지는 것을 알 수 있었다. 즉, 발효한 산양유를 섭취함으로 외부로부터 침입한 병원균이나 항원물질에 대해 면역세포의 반응성이 증가하여 생체의 저항력이 향상되는 것을 기대할 수 있다. 또 다른 선행연구에서는 산양유에 Lactobacillus rhamnosus 균주를 접종하여 만든 발효유를 투여함으로써 영양실조 상태가 회복되고, 장과 호흡기 감염에 대해 저항력이 높아진다고 보고되었다(Salva et al., 2011). 그러나 산양 발효유의 생리활성에 대한 연구는 아직 부족하여 향후 더 많은 연구가 필요한 실정이다.

산양유는 다른 유제품보다 많은 비율의 올리고당을 함유하고 있다(Kim et al., 2014). 산양유에 포함된 올리고당은 6-sialyl-lactose, 3-sialyl-lactose, N-glycolylneuraminyl-lactose, 3-galactosyl- lactose, N-acetylglucosaminyl-lactose 및 높은 분자량을 가진 올리고당으로 모유와 유사한 조성을 가진다(Martinez-Ferez et al., 2006; Silanikove et al., 2010). 산양유의 올리고당은 프리바이오티스(prebiotics)의 속성을 갖고 있어, 인체에 이로운 영향을 미치는 Bifidobacterium과 젖산균(Lactobacillus 등)과 같은 프로바이오틱스(probiotics) 균주들이 이용할 수 있으며, 유기산과 단쇄지방산(short chain fatty acid)을 생성시켜, 장내의 pH을 조절함으로 산에 민감하고 식중독의 원인균으로 알려진 Clostridium perfringens 균주 등과 같은 유해균을 억제시킨다(Kleessen et al., 1997; Fooks et al., 1999; Kwon and Lee, 2002).

Choi 등(2015)의 연구에서도 산양유 섭취를 통해 올리고당 성분이 성인의 분변 내 비피도 박테리아의 증식 효과 및 당 발효 능력을 조사하였으며, 산양유를 섭취한 처리군이 대조군에 비해 비피도박테리아의 균수가 증가한다고 보고하였다.

4. 산양유의 생산 및 상업적 활용

산양유와 관련된 유제품 소비에 따른 생산량은 나날이 증가하는 추세이며, 여러 나라의 산양유제품 산업에 관한 수치는 Table 7에 제시하였다. 유럽 국가 중 특히 프랑스에서 산양유 산업이 활발하게 육성되고 있다. 특히 산양유는 지중해, 중동, 동유럽 및 남미국가와 같은 많은 개발도상국에서 이미 다양한 가공을 통하여 소비되고 있는 반면, 아직 우리나라에서는 그 시장이 크지 않다.

우리나라에 1903년 Saanen종이 처음 도입되었지만, 실질적으로 1960년대 초부터 알려지기 시작하였으며, 2008년도에는 전국에 약 5,800여 두의 유산양이 충북 영동 및 강원도 홍천을 중심으로 사육되었으며, 평균 1일 약 5톤의 산양유가 생산되었다(Ahn and Park, 2008). 산양은 반추동물 중에서도 크기가 작고 민첩하여 우리나라 지형에서 사육하기 적합할 뿐만 아니라, 질병에 강하고 젖소보다 체중 대비 산유량이 2배나 높은 특성을 지녀 산업적 이용가치가 큰 가축이다(Parkash and Jenness, 1968; Ahn and Park, 2008).

산양유의 가장 대표적인 활용 방안은 기본적으로 영양 취약계층이나 일부 알레르기 등으로 충분한 영양섭취가 이뤄지지 않는 어린이나 노인들에게 적합한 식품원이다.

산양유를 상품화하기 위해 가장 고려해야 하는 부분은 특유의 냄새와 독특한 풍미를 거부감 없이 받아드릴 수 있도록 가공하는 것이다. 특히 산양유의 지방성분은 산양유의 색, 향, 치즈의 경도 등에 영향을 미치면서, 잠재적으로 인간 소비자의 건강에 긍정적이거나 부정적인 요소로 작용할 수 있는 주요 요인으로 알려져 있다(Ribeiro and Ribeiro, 2010). 영국에서는 많은 산양유의 장점에도 불구하고, 부정적인 인식으로 인한 소비 감소를 개선하고자 의료제품, 기능성 식품, 치즈, 아이스크림 등 다양한 방법으로 소비자에게 제공되고 있으며, 그 결과 연간 15~20만 리터의 산양유가 소비되고 있다. 산양유 유제품 중 60%는 치즈, 20%는 액상유, 10%는 발효유로, 나머지는 버터, 크림, 아이스크림으로 섭취되고 있다(Mowlem, 2005).

산양유를 상품화 하기 위해 고려해야 할 또 다른 특성은 우유와 다른 케이신 마이셀의 크기와 구조이며, 단백질 분해 효소의 작용에 차이를 보여 제품화 공정에 영향을 미친다는 것이다(Pierre et al., 1995; Bramanti et al., 2003). 또한 제품의 신뢰성과 규격화된 평가방법이 정립되어야 하고, 열처리에 많은 영향을 받는 유청 단백질의 변성을 최소화하는 방법을 강구하여야 한다(Bramanti et al., 2003).

많은 연구를 통하여 산양유의 생리 활성에 미치는 다양한 이로운 점을 부각시켜 기능성 식품 시장을 형성하고, 향후 일반 시장에도 치즈, 발효유, 과자, 버터 등의 유제품 제조를 통해 산양유의 활용성을 높여 나간다면 그 수요는 증가할 것으로 판단된다.

결 론

산양유는 총고형분, 단백질, 지방, 무기질 성분이 적절히 함유되어 있고, 유당의 함량은 낮아 영유아 및 노인의 영양공급원으로 적합하다. 산양유의 주요 단백질 조성은 모유와 유사하여 β-케이신 함량이 높기 때문에 부드러운 커드를 형성하여 소화 흡수에 용이하다. 특히 산양유의 β-케이신은 대부분 A2 변이체로 존재하여 다양한 잠재적인 질병 요인과는 무관한 것으로 보고되었다. 산양유에는 필수 아미노산이 고르게 함유되어 있고, 카프릴산과 카프릭산 등의 중간사슬지방산의 함량이 높아 지방 조직에 축적되지 않고 바로 에너지로 사용되거나, 체내에 콜레스테롤이 축적되는 것을 방지하여 다양한 질환의 치료제로 각광받고 있다. 또한 산양유는 비타민과 무기질의 함량이 풍부하고, 칼슘, 인, 망간의 체내 흡수율 및 이용률이 뛰어나다.

이러한 산양유의 영양성분 조성과 함량은 산양의 사료조건, 계절, 지역, 건강상태 등의 조건에 따라 변화될 수 있기 때문에 고품질의 산양유를 생산할 수 있는 사육 조건에 대한 연구가 필요하다. 산양유는 건강 유지 및 생리적 기능 개선에 이로운 효과가 있고, 특히 소화 흡수율이 뛰어나 영유아 및 노인들의 좋은 영양공급원이 될 수 있음에도 불구하고, 우유에 비해 국내 산양유 시장의 규모는 매우 작을 뿐만 아니라 산양유의 영양학적 우수성이 일반 소비자들에게 정확히 전달되지 못하고 있기 때문에 산양유의 영양학적 가치에 대한 정보 제공이 필요하다.

더불어 다양한 나라에서 소비되는 산양유의 방안을 살펴 우리나라 실정에 알맞게 적용한다면 산양유의 유익한 특성이 필요로 되는 대상자들에게 적절히 적용될 것이다. 또한, 산양유를 이용한 식품 제조는 고령화 사회에 접어드는 우리나라에서 그 활용도를 상승시킬 수 있어 앞으로 산양유 제품의 기능성에 대한 체계적인 연구가 더욱 활성화 되어야 할 것이다.
